Chlormadinone acetate (Gestafortin) is a steroidal progesterone with anti-androgenic and anti-gonadotropic effects. Its half-life is approximately 34-38 hours.

Production Method
In a 2000mL three-necked flask, add 100g of self-prepared chlormadinone acetate and 1000mL of glacial acetic acid. While stirring, add 50g of acetic anhydride and 2g of p-toluenesulfonic acid. Slowly heat the reaction mixture and stir at 40-45℃ for 8-12 hours. After the reaction is complete, add 10mL of 30% sodium hydroxide solution to neutralize the strong acid in the reaction solution. Then concentrate under reduced pressure to recover 85-90% of the glacial acetic acid. Add 600mL of tap water to the concentrated reaction solution, cool to 10-15℃, and stir for 2-3 hours to promote crystallization. The crystals were collected by filtration, washed with water until neutral, and the filter cake was dried below 70°C to obtain 114.2 g of crude product (8R,9S,10R,13S,14S,17R)-17-acetyl-6-Chemicalbookchloro-10,13-dimethyl-3-oxo-2,3,8,9,10,11,12,13,14,15,16,17-dodecano-1H-cyclopentano[a]phenanthrene-17-yl acetate, with an HPLC purity of 98.8%. The filtrate and washings were combined, and the residual solvent was recovered and disposed of to the wastewater treatment system. The crude product was decolorized and recrystallized using conventional methods with alcohol and activated carbon to obtain 97.8 g of pure product with a melting point of 208-212°C, an HPLC purity of 99.5%, and a yield of 97.8%.
