Introduction
Indomethacin, chemically known as 2-methyl-1-(4-chlorobenzoyl)-5-methoxy-1H-indole-3-acetic acid, belongs to BCS Class II drugs. It has high permeability, low solubility, poor dissolution rate, and low bioavailability in vivo. Indomethacin is a nonsteroidal anti-inflammatory drug (NSAID) with anti-inflammatory, antipyretic, and analgesic effects. Its anti-inflammatory effect is achieved by inhibiting cyclooxygenase, reducing prostaglandin synthesis, while its antipyretic effect is related to the inhibition of prostaglandin synthesis in the hypothalamus.
Chemical Properties
White or slightly yellow crystalline powder. Melting point 158-162℃. Soluble in acetone, slightly soluble in ethanol, chloroform, and ether, almost insoluble in water. Tasteless and almost odorless.
Uses
Indomethacin has significant anti-inflammatory and antipyretic effects, mainly used for rheumatic arthritis, ankylosing spondylitis, osteoarthritis, etc., in patients who are not tolerated or do not respond well to salicylates.
Used as a non-hormonal anti-inflammatory and analgesic.
Uses: Cyclooxygenase (COX) inhibitor, selectively inhibiting COX-1; blocking the biosynthesis of prostaglandins. Pharmacologically, it has significant anti-inflammatory and antipyretic effects, mainly used for rheumatic arthritis, ankylosing spondylitis, osteoarthritis, etc., where patients are not well tolerated or do not respond well to salicylates. Production method: p-Aminoanisole is obtained through diazotization, reduction, acidification, hydrolysis, and cyclization to produce indomethacin.
