Overview: Memantine hydrochloride is a dementia treatment drug developed by Merz AG in Germany. It is a novel, low-to-moderate affinity, voltage-dependent, non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist. It non-competitively blocks NMDA receptors, reduces glutamate-induced NMDA receptor over-excitation, prevents apoptosis, and improves memory. It is a new generation of drugs for improving cognitive function. In February 2002, the European Committee on Patents for Medicinal Products (CPMP) approved it for the treatment of moderate to severe Alzheimers disease. It was launched in Germany in August of the same year, and on October 17, 2003, it was approved by the U.S. Food and Drug Administration (FDA) for the treatment of moderate to severe Alzheimers disease. Further research showed that memantine hydrochloride is also effective for mild to moderate Alzheimers disease. Alzheimers disease (AD), also known as senile dementia, is one of the most common diseases among the elderly, mainly characterized by memory loss and cognitive impairment. It is a progressive neurodegenerative disease. Currently, cholinesterase inhibitors (donepezil, rivastigmine, galantamine) and memantine hydrochloride are approved for the treatment of Alzheimers disease.
